THE COMPLETE AMINO ACID SEQUENCE OF THE a-LACTALBUMIN FROM A MARSUPIAL
نویسنده
چکیده
a-Lactalbumin was purified from a whey protein fraction of the milk of the red-necked wallaby (Macropus rufogrisem). The complete amino acid sequence was determined from the results of automatic sequenator analyses of the intact protein, the three cyanogen bromide fragments, and of peptides generated from the larger, COOH-terminal CNBr fragment by digestion with trypsin or staphylococcal protease. This is the first sequence to be determined of an a-lactalbumin from a marsupial and iffers from known eutherian alactalbumins in size and locations of deletions in alignments with the homologous type c lysozymes, as well as in having amino acid substitutions at 8 sites that are invariant in known eutherian proteins. Some corrections are also reported for two regions of sequence in both bovine and goat a-lactalbumins. The new and previously published information on a-lactalbumin sequences is analyzed in relation to the evolutionary history of the a-lactalbumin line as well as the relationship of structure to function in these proteins.

The Complete Amino Acid Sequence of Bovine a-Lactalbumin*
The complete amino acid sequence of bovine oc-lactalbumin has been established. The unique seq, uence was deduced by characterization of the tryptic, chymotryptic, and peptic peptides isolated from enzymic hydrolysates of the two unique fragments obtained on cleavage of S-aminoethyl-a-lactalbumin with cyanogen bromide as described earlier. Important overlapping peptides were also obtained by seq...
متن کاملMolecular characterization of apolipoprotein A-I from the skin mucosa of Cyprinus carpio
Apolipoprotein A-I is the most abundant protein in Cyprinus carpio plasma that plays an important role in lipid transport and protection of the skin by means of its antimicrobial activity. A 527 bp cDNA fragment encoding C terminus part of apoA-I from the skin mucosa of common carp was isolated using RT-PCR. After GenBank database searching, a partial sequence containing a coding sequence (CDS)...

متن کاملThe construction, identification and characterisation of plasmids containing human alpha-lactalbumin cDNA sequences
We describe the cloning of double-stranded cDNA synthesized from lactating human mammary gland total poly(A)-containing RNA, into the EcoRI site of the plasmid pAT153. Nine recombinants were shown to contain alpha-lactalbumin cDNA sequences as determined by positive hybridisation translation of complementary RNA. Restriction enzyme maps were determined for six of these. Alignment of the restric...
متن کاملNucleotide sequence of cDNA encoding for preprochymosin in native goat (Capra hircus) from Iran
Prochymosin is one of the most important aspartic proteinases used as a milk-clotting enzyme in cheese production. In the present investigation we report sequence of cDNA encoding goat ( Capra hircus ) preprochymosin and compare its nucleotide and deduced amino acid sequences with sequences of other ruminants preprochymosin. As bovine prochymosin, the caprine prochymosin cDNA encodes 365 amino ...
